Two questions what's the best way of sending Tyres interstate ? Aussie post....
Is anyone heading up from melb to sydney before Tuesday of next week and would have room for a front and rear tyre?

Australia Post is the go. Don't bother boxing them up, just tape the tyres together (so they can't be separated!) and send them like that. I think it cost around $20-25 to send two from here to Melbourne when I did it.
